Talking about spin-offs and/or adaptations of Homestuck into other media, I think the Condescension era of B2 Earth could actually be really great to explore in a live-action movie.
Not much Homestuck-related info needs to be provided there because it would take place on Earth, and the only characters who'd be in both Homestuck and in this hypothetical movie would be the Condesce and possibly B2 Dave and B2 Rose (plus ICP and Guy Fieri, maybe?). And if some of the Condesce's motives get explained in a way like Dirk explained them to Jake, that would be enough background info probably to work with.
I think a movie could be a really good medium to explore that timeframe more.

Remember, though, adapting a work into a different medium is usually always going to be hard. Besides Comic > Movie, it's usually going to have to change a bunch. But if done well, the spirit of the source material can still be kept. There was a Radio Drama adaptation of Star Wars in the early 1980s, and it was actually pretty phenomenal somehow. I'm sure Homestuck could do the same, though it'd have to alter the plot quite a lot. It'd be about the same as a cartoon version in terms of changes, except that a Radio show would have to move at a slower pace and would lose many of the visual jokes that the cartoon version would be able to keep.

It could always a be a horror-comedy movie. ;o) Or you leave out mentions of the ICP, Guy Fieri, and SBAHJ. Or heck, just let the movie take place AFTER the ICP and Guy Fieri have already been killed and Dave and Rose have been taken out by the Condesce as well already. Then you got mostly very serious material to work with.

Nah I think the humor is an integral part of that backstory. A retelling of Davd and Momxy's media war against the Presidents and Condesce would have a sad ending and a lot of Jokes, but would have actual recognizably Homestuck characters. Something taking place after their deaths would not really be Homestuck, IMO.

There is a very strong precedent for comical/video-gamey source material surviving the adaptation to film... I am, of course, referring to the one-hit wonder Scott Pilgrim v. The World. It's got video game abstractions slapped right on top of the real world, blatant CG flairs, and it's unapologetic about it... and people still loved it.
Do I think Homestuck could be adapted faithfully to cinema? Absolutely. Do I think it's likely? No. There are so many barriers to delivering a product that's artistically distinct and distinguishable. The movie-going populace tends to like a handful of proven motifs and messages, delivered through variably themed settings - that is, the same cake with different icing. There's nothing wrong with that, but it does naturally incline the industry to favor the creation of less adventurous works.
Because of that, all the shot-callers involved with production will get their words in edgewise, and so the nature of the story is changed in the process. Broadly speaking, what you started out wanting to recreate is not what is created. It... pains me to cite an example, but I will point to director M. Night Shyamalan's cinematic adaptation of Avatar: The Last Airbender. Absolutely phenomenal source material. Critically acclaimed and loved by audiences. Movie adaptation? 6% on Rotten Tomatoes.
I don't pretend to know that the tragedy is a result of Shyamalan trying to make the material his own or if it was a product of executive meddling - I have read reports of both - but I do know that the same risk factors are present no matter what story people are trying to remake. I'll keep my hopes up, though.
EDIT: In line with the Scott Pilgrim comparison, I do think it could be done in live-action. Especially if it's CG-assisted, like the other Avatar, James Cameron's more recent magnum opus.

Obviously, it's not perfect since it's limited to existing film clips, but I always thought it captured the "feel" of what the (first) Homestuck movie may be like. Like I've said before, were it done right, I'd love to see a series of Homestuck live-action flicks.

It should be noted that John was able to retcon teleport into Con Air as it was played on a TV. Unless he simply teleported into the set, John's retcons extend outside the bounds of even paradox space itself. Unstuck in existence, if you will.

Everything he teleported into was previously shown in The Comic, if memory serves. Con Air works because he and Jade had been watching it at the time. What would've been weirder would have been if he entered e.g. a Paradox Space comic, or yeah the real world.

Personally, I think that having Hollywood actors and directors creating the movie would sorta ruin Homestuck in a way. If Homestuck were to have a film/tv adaptation, it should be produced/acted/written by actual fans or contributors to the comic itself. Imagine class-act cosplayers and roleplayers playing as Homestuck characters in an actual production! The amount of love for the characters portrayed by actual readers of the comic would show probably more than if Brad Pitt was acting...
I think that if it was done right, Homestuck could become an AMAZING film/tv show, but there is so much room for error and so many ways it could screw up.

As much as I really want to be excited for a film, I just don't think Hollywood would give it justice unfortunately...

IMO, when it comes to flim/movie adaptations of Homestuck I think that something animated would be better than something live-action, if only because it would be easier and match the source material even more. Scott Pilgrim works because it takes place in our universe, just framed from a comic book's viewpoint. Homestuck takes place on mostly alien planets with bizzare landscape. Not to mention the fact that something like Homestuck would most likely be rated R for language alone (although that could be censored for mass appeal.)
I also think something like a TV show would work better for the whole premise because it can allow the story to develope a lot better. Not to mention the fact that animated TV shows are popular with the HS demongraphic anyway, so it'll be a perfect fit. Most animated movies are more family-oriented, which makes it harder to produce something like Homestuck into a movie format unless it's live-action.
